Transcription factors (TFs) are a group of cellular proteins that play essential roles in regulating gene expression. Monitoring the activation of multiple TFs simultaneously is critical to understand the molecular mechanism of cellular regulation underlying cell signaling and gene expression. Signosis’ Combo TF activation profiling plate array is a single array that can be used for the measurement of the activity of 192 different TFs containing comprehensive functional TFs for ER stress, mitochondria UPR stress, Oxidative stress, as well as disease-related TFs, including heart, liver, kidney, ovary, etc.  Optimal for analysis of multiple mammalian cells such as human, mouse, rat, sheep, and pig etc. The differences of samples can be quantitatively analyzed and compared at once. There are four 96-well plates included in the kit along with the reagents for the assay.

Principle

Signosis’ TF Activation Profiling Arrays are used for monitoring the activities of multiple TFs simultaneously. In the technology, a series of biotin-labeled probes are made based on the consensus sequences of TF DNA-binding sites. When the probe mixtures incubate with nuclear extracts, individual probes will find their corresponding TFs and form TF/probe complexes, which can be easily separated from free probes through a simple spin column purification. The bound probes are then detached from the complexes and analyzed through hybridization with a plate, which each well is specifically pre-coated with complementary sequences of the probes.
