The plenti-EGFR (viii) vector contains the mutation EGFR viii under the MSCV promoter which can be used to infect both dividing and nondividing cells. It contains the HIV-1 LTRs and the lentiviral packaging signal (Ψ) for retroviral expression, with specific elements (WPRE, cPPT, RRE) included for improved transgene expression. Downstream of the EGFR viii gene is a DYKDDDDK-tag for Western blotting. Furthermore, copGFP can be used as a selection marker when used with stable cell lines.

Epidermal growth factor receptor (EGFR) is a transmembrane tyrosine kinase receptor that plays a critical role in regulating cell division and death.  The EGFR viii mutation (also known as EGFR variant III, de2-7EGFR, and ΔEGFR) is one of the most common mutations found in human cancers. This mutation results in the overexpression of EGFR with an in-frame deletion of 801 base pairs representing exons 2-7. This deletion removes 267 amino acids and results in a significantly shorter extracellular domain that is unable to bind to any known ligand.
