The miR-17-92 cluster, containing miR-17, miR-18a, miR-19a, miR-20a, miR-19b-1 and miR-92a-1, is among the best-studied microRNA clusters. Over-expression of the miR-17-92 cluster is a key oncogenic event in various cancer types, which are involved in many oncogenic signaling pathways, PTEN, TGFβ signaling, Notch and Sonic Hedgehog pathway. Research work in recent years has revealed unexpected roles of miRNA-17-92 cluster due to its members have function in a wide variety of settings that include normal development, immune diseases, cardiovascular diseases, neurodegenerative diseases and aging. Signosis has developed miR-17-92 cluster real-time PCR kit to detect the expression of miR-17-92 simultaneously in a high sensitive and discriminative manner. One kit allows 12 reactions.
Principle
In the assay, the target miRNA molecule is hybridized with two oligos to form a RNA/DNA duplex. When the sequences are perfectly matched, they are aligned with the miRNA and the joint can be ligated with DNA ligase. A single nucleotide difference among miRNAs will block either the hybridization or the ligation. After the pair of oligos is ligated, the ligated molecules are subjected to real-time PCR analysis.
