The plenti-EGFR (A767dupASV, C797S) vector contains EGFR with the A767dupASV, C797S mutations under the MSCV promoter which can be used to infect both dividing and nondividing cells. It contains the HIV-1 LTRs and the lentiviral packaging signal (Ψ) for retroviral expression, with specific elements (WPRE, cPPT, RRE) included for improved transgene expression. Downstream of the EGFR viii gene is a DYKDDDDK-tag for Western blotting. Furthermore, copGFP can be used as a selection marker when used with stable cell lines.
Epidermal growth factor receptor (EGFR) is a transmembrane tyrosine kinase receptor that plays a critical role in regulating cell division and death. The EGFR A767dupASV, C797S mutations include both a duplication of the ASV amino acids (Ala, Ser, Val) at position 767 (in the α-C helix), and a point mutation that changes a C (Cys) into an S (Ser) at point 797. The C797S mutation in particular is known to confer resistance to EGFR inhibitors such as osimertinib and poziotinib.
