The plenti-EGFR (wt) vector contains the wt EGFR gene under the MSCV promoter which can be used to infect both dividing and nondividing cells. It contains the HIV-1 LTRs and the lentiviral packaging signal (Ψ) for retroviral expression, with specific elements (WPRE, cPPT, RRE) included for improved transgene expression. Downstream of the EGFR viii gene is a DYKDDDDK-tag for Western blotting. Furthermore, copGFP can be used as a selection marker when used with stable cell lines.
Epidermal growth factor receptor (EGFR) is a transmembrane tyrosine kinase receptor that plays a critical role in regulating cell division and death. The EGFR gene is present on chromosome 7p11.2 and has 28 coding exons in which exons 5–7 and 13–16 code for the ligand binding domain and exons 18–24 code for the TK domain. Downstream effects of EGFR activation include modulation of three major pathways of RAS-RAF-MAPK, PI3K/AKT and JAK/STAT pathways which can lead to cell survival, proliferation and migration of cancer cells.
