The pMSCV-EGFR (D770_N771insSVD, C797S) vector contains EGFR with the D770_N771insSVD, C797S mutations under the MSCV promoter for high expression in hematopoietic, HEK293, and stem cells. Downstream of the mutant EGFR gene is a DYKDDDDK-tag for Western blotting. Furthermore, the coGFP under the EF1 promoter can be used as a selection marker with stable cell lines. The base vector is a modified pMSCV vector with the LTRs and the lentiviral packaging signal (Ψ) removed for standard expression. Epidermal growth factor receptor (EGFR) is a transmembrane tyrosine kinase receptor that plays a critical role in regulating cell division and death. The EGFR D770_N771insSVD, C797S mutations involve the insertion of amino acids SVD (Ser, Val, Asp) between the position D770 and N771, and a point mutation that changes a C (Cys) into an S (Ser) at point 797. The C797S mutation in particular is known to confer resistance to EGFR inhibitors such as osimertinib and poziotinib.
