Signaling pathway is to convert extracellular signals, whether it is cytokine, insulin, or antigen, into intracellular changes with the same end results: alteration in transcription activity, resulting in changes in transcription factor-binding patterns at gene regulatory regions. Classically, the electrophoretic mobility shift assay (gel shift) and its based array assays, have been the assay of choice for demonstrating that a protein family can bind to a specific consensus sequence. The assays can only detect the activity of the TF gene family but cannot distinguish individual gene members. The individual transcription factors always play distinctive important roles in different signal pathways such as Smad 1 in BMP pathway and Smad3 in TGFbeta pathway.  In order to pinpoint the functions of individual TFs, Signosis provides QuantiTF ELISA kits for quantitatively analyzing DNA binding activities of a specific TF with an antibody against the TF and the corresponding active recombinant protein as a positive and potential standard.
